A citometria de fluxo é um método útil para obter o fenótipo e as caraterÃsticas das células. Baseia-se na dispersão da luz pelas células em estudo e na emissão de fluorescência pelas mesmas. A emissão de fluorescência é obtida através da utilização direta de corantes fluorescentes ou de uma combinação de corantes fluorescentes e anticorpos monoclonais. Estes anticorpos conjugados com fluorescência podem detetar e ligar-se a moléculas de superfÃcie ou a compostos internos das células, tornando possÃvel a identificação de tipos de células numa população celular diversificada por citometria de fluxo. Este método foi basicamente desenvolvido por imunologistas que tentaram separar populações puras de células umas das outras e, após a sua proliferação em meio de cultura celular, estudar o papel individual de cada célula no sistema imunitário. Atualmente, a utilização de tecnologias modernas para aumentar a velocidade e a facilidade de execução dos trabalhos está a expandir-se cada vez mais. A citometria de fluxo é normalmente utilizada para identificar linhas celulares, analisar tecidos celulares maduros e detetar a heterogeneidade da população de células tumorais. Na última década, a utilização deste método em laboratórios clÃnicos e no diagnóstico de vários tipos de cancro aumentou drasticamente.
